Human Genome Project - Mapping

More info : Completed in 2003, the Human Genome Project (HGP) was a 13-year project coordinated by the U.S. Department of Energy and the National Institutes of Health. During the early years of the HGP, the Wellcome Trust (U.K.) became a major partner; additional contributions came from Japan, France, Germany, China, and others. Project goals were to identify all the approximately 20,000-25,000 genes in human DNA, determine the sequences of the 3 billion chemical base pairs that make up human DNA, store this information in databases, improve tools for data analysis, transfer related technologies to the private sector, and address the ethical, legal, and social issues (ELSI) that may arise from the project.

The process of making a gene library
The process of making a gene library The DNA molecules of an organism in interest are isolated. The DNA molecules are then partially digested by a endonuclease restriction enzyme. Sometimes, the DNA molecules are digested to different lengths of time in order to ensure that all the genes have been digested to manageable sizes. The digested DNA molecules are run on agarose electrophoresis for which a suitable range of lengths of DNA pieces are isolated and ligated to vector plasmids. Statistically, 99% of the genes will be encoporated into the plasmids. The plasmids then can be taken up by suitable hosts. The hosts are kept in liquid media and can be frozen at -80oC for a long time. The process of subdividing genomic DNA into clonable element and inserting them into host is called creating a library, a clone bank or a gene bank. A complete library of host cell will contain all of the genomic DNA of the source organism
